Psilocybin and SNRIs
Reduces effectsAntidepressant, SNRI · also: venlafaxine, effexor, efexor, desvenlafaxine, duloxetine
SNRIs such as venlafaxine and duloxetine tend to weaken mushroom effects, similar to SSRIs, and the main risk is redosing.
Blunted effects, as with SSRIs. Venlafaxine carries a little more risk than the rest of the class.

Why it happens
SNRIs block reuptake of both serotonin and noradrenaline, and the blunting is thought to work as with SSRIs, through adaptation of 5-HT2A receptors over weeks. Psilocin is a direct receptor agonist, so a classic serotonin-release interaction is not expected. Venlafaxine has a separate seizure warning: seizures have been reported during treatment and are among the more commonly reported events in overdose.
What is known
By extension from the SSRI data plus survey reports, in which SNRIs showed a similar weakening. Less directly studied.
- 2023
- In an online survey, the estimated probability of weaker than expected mushroom effects with an SNRI was 0.55, similar to SSRIs (0.47). Gukasyan et al., Journal of Psychopharmacology 2023
- 2022
- A systematic review found no controlled psilocybin study with an SNRI; the only controlled reuptake-inhibitor study with psilocybin used escitalopram, while duloxetine was studied with MDMA, where it broadly attenuated effects. Sarparast et al., Psychopharmacology 2022 (systematic review)
- 2025
- The Effexor XR label states that seizures have been reported with venlafaxine and advises caution in people with a seizure disorder; seizures are listed among the most commonly reported events in postmarketing overdose. EFFEXOR XR (venlafaxine) US prescribing information, DailyMed
- 2025
- The Effexor XR label recommends gradual dose reduction rather than abrupt cessation because of discontinuation symptoms, which can include seizures. EFFEXOR XR (venlafaxine) US prescribing information, DailyMed
Safety guidance
- Expect a weaker experience and avoid redosing during the come-up.
- Venlafaxine is the one in this class to be more careful with because of its seizure warning, especially at high doses.
- Do not stop or skip an SNRI to take mushrooms; venlafaxine in particular needs a gradual taper agreed with your prescriber.
- Avoid adding other serotonergic or seizure-threshold-lowering substances on the same day.
When to seek help
Call emergency services for a seizure, or for confusion with fever, rigidity, muscle jerking or a racing heart. Emergency red flags.
Common questions
Do SNRIs blunt mushrooms like SSRIs?
Survey data suggest yes, at a similar or slightly higher rate (0.55 vs 0.47 probability of weaker effects).
Why is venlafaxine singled out?
Its label carries a seizure warning and seizures feature in overdose reports, so it adds a risk the other drugs in the class are less associated with.
Can I skip my dose the day before?
Missed doses can bring on discontinuation symptoms, and the label advises gradual changes. Talk to your prescriber rather than adjusting it yourself.
