Psilocybin and trazodone
Reduces effectsAntidepressant, SARI · also: desyrel, molipaxin, oleptro
Trazodone, even at low sleep doses, is expected to blunt a mushroom experience and add sedation, though the pair has not been studied.
Blunts the experience and adds heavy sedation.

Why it happens
Trazodone has dose-dependent actions: at low hypnotic doses it blocks 5-HT2A, histamine H1 and alpha-1 adrenergic receptors, and at higher doses it also blocks the serotonin transporter. Because psilocybin's effects depend on 5-HT2A activation, a 5-HT2A antagonist is expected to reduce them. Its label also carries a general serotonin syndrome warning when combined with other serotonergic drugs.
What is known
Based on the mechanism, plus consistent report. Not formally studied with psilocybin.
- 2009
- Trazodone is hypnotic at low doses through blockade of 5-HT2A, H1 and alpha-1 receptors; higher doses add serotonin transporter blockade. Stahl, CNS Spectrums 2009 (abstract via publisher record)
- 2026
- The US label describes trazodone as both a serotonin reuptake inhibitor and a 5-HT2 receptor antagonist and warns of increased serotonin syndrome risk with other serotonergic agents. Trazodone hydrochloride tablet US prescribing information, DailyMed
- 1998
- In healthy volunteers, psilocybin's effects were blocked dose-dependently by the 5-HT2A antagonist ketanserin. Vollenweider et al., NeuroReport 1998
Safety guidance
- People often forget that a sleep dose of trazodone is still a 5-HT2A blocker; expect weaker effects.
- Do not redose to overcome the blunting.
- Expect added drowsiness, and avoid driving or being alone near hazards.
- Do not change your trazodone dose for a trip without your prescriber.
When to seek help
Get urgent help for fainting, a very slow or irregular heartbeat, extreme drowsiness, or confusion with fever, sweating and muscle jerking. Emergency red flags.
Common questions
I only take trazodone for sleep. Does it matter?
Yes. Low sleep doses are exactly where its 5-HT2A blocking action is most prominent, so mushrooms may feel weaker.
Is serotonin syndrome a risk?
The trazodone label warns about it with serotonergic drugs in general. With psilocybin it is not documented, and psilocin does not release serotonin.
Should I skip trazodone the night before?
Discuss any change with your prescriber; it has not been studied with psilocybin.
